Development by Hitachi that might go into the Roche clinical MS: "These results suggest that the 8–4 pole ion guide achieves higher transmission efficiency relative to the conventional quadrupole ion guide under a high inlet gas flow rate."

Rumour is that Sciex are currently the lowest priced top-tier MS right now, with the 8600 coming in around $1 million USD. They should make their money back in other ways (software subscriptions in particular), but if you want the cheapest, single cell-level MS, the 8600 is probably it.

If anyone was curious. We succeeded, but it wasn’t easy. Reviewers were insistent on western blots. Eventually we convinced the editors otherwise using peptide level quant info to show decent sequence coverage and a biochemical/biophysical rational for why not all the peptides had the same quant.

In case you missed it: DNA miniprep columns are an equivalent replacement for the classic proteomics suspension-traps, and 10x cheaper. $5 a sample in prep just doesn't scale so it's either SP3 or this approach. pubs.rsc.org/en/content/a...

Friends don’t let friends identify glycopeptides by deamidation. Even non-glyco proteomics people know that deamidation can be an artifact. Either intact or endoglycosidase (see Yates lab DeGlypher approach) is best.

The University of Michigan now blocks the iProX database, which is a part of the PRIDE consortium of mass spec data repositories. All requests to unblock were denied. Any other US universities in a similar situation? There is a lot of valuable MS proteomics data there no longer accessible to us.

In summary, we’ve found the rules to understand what a site needs to be glycosylated - an Asn in an NxS/T sequon without secondary structure and with a local sequence that tightly binds the OST groove to switch the enzyme ON.
