Happy to share this paper! We uncovered that good 5' splice sites protect against ZC3H4/Restrictor's promiscuous role, and that its role is to suppress elongation and indirectly cause termination. Looking forward to learning more about this protein in the future!

We have had multiple discussions about why people have not previously observed the very dramatic reduction in transcription upon PNUTS depletion. So I looked at our own TT-seq data and analysed it without including the spike-in. The numbers speak for themselves! Spike-in calibration is important!

Final note: The success of this project relied on collaboration. We couldn’t have done it without bringing together different approaches, from genomics and proteomics to biochemistry to live-cell imaging. Every author put in a lot of hard work and I’m so proud to share credit with them all (8/8)

A project that briefly dragged us away from CpG islands before leading us back to the 5’ end of genes!

A preview on the latest work from @robklose.bsky.social lab where Aleks (Not made the bluesky transition yet) used single molecule approaches to address how Polycomb controls transcription. He found that Polycomb limits the frequency with which a promoter can enter a permissive transcribing state.
