It was a good experience to step back and briefly take stock of the amazing progress in connectomics since I started working on this stuff (20 years ago!) thanks as well to @natrevneuro.nature.com for the constructive editorial interactions.

X-ray nanotomography can be your new friend for neuronal imaging. In our new #preprint (tinyurl.com/nanoXneuro) we present advances that push the spatial resolution limit for X-ray holographic nanotomography (XNH). We can now resolve synapses. 🧵

Couldn't agree more. There’s still so much to be uncovered in the dataset. One thing that wasn’t emphasized before is how it enables access to specific circuit motifs down to identified cells. 12 examples are highlighted in the preprint, but again just the tip of the iceberg.

Our fly brain connectome papers are now live (www.nature.com/immersive/d4...), and is getting traction (www.bbc.co.uk/news/article...). With ~139k neurons, >8k cell types, synapses annotated, transmitters predicted, ~134 evo-devo units denoted, so much is now possible in the fly.

The fly now has a full #connectome, in its most basic form a directed + weighted graph. But what are its connection ‘signs’, excitatory/inhibitory/modulatory? Our new, now published work moves towards this answer, predicting #neurotransmitters from EM! 1/n www.cell.com/cell/fulltex...

Good morning #SfN23 people! Our lab has 3 posters this morning (Sunday Nov 12) and 2 tomorrow morning (Monday Nov 13). We'd love to see you and share our latest on fly vision, circuits, and behavior.

A neat technical aspect of this study is that we went back to the same TEM grids imaged a decade earlier and re-imaged them at sub-nanometer resolution for additional information. A distinct advantage of sample-preserving EM and as Sydney Brenner noted data that is complete, accurate, and permanent.

Ran a nice section of the Sarapiquí with an outfitter last week. A near continuous 2-hour stretch of class III/IV with nice boilers and surfing holes, interspersed with a couple 90 degree walls and some technical drops. Felt like a cross between the Dead and Penobscot rivers. Pura Vida!

Food at local CR sodas was great. I was particularly taken with delicacy of tamales at El Turnito. Each soda seems to have its own delicious chilera recipe to boot.

Heading home from a week in Costa Rica. Amazing flora and fauna. And water, lots and lots of water.
