Tuberous sclerosis complex is a genetic disorder associated with neurological symptoms that have been attributed to mTOR signaling. In this work, Afshar-Saber et al. demonstrate that altered mTOR activity disrupts neuronal differentiation and changes in DNA methylation. This results in rapamycin, an mTOR inhibitor, being ineffective in rescuing neuronal hyperactivity after maturation.
