Mammalian systems, how stable are #fluorescent protein fusions in living mammalian cells? It used to be that transfection of epithelial cells were ~20% successfull in a monolayer of cells, which to me was unacceptable. Has this changed?

1-2μm/°C is a good 'rule of thumb' for the axial shift of the focal plane of an objective with temperature, or ~1-2 depths of field (DOF) at high numerical aperture. -> Here's a typical lab trace with a swing of 1-2°C over 1-2h. This is ~1-4 DOF of drift per hour! So continuous autofocus is needed!
