Ignorant drug development question. What’s missing on target-based drug development to make it truly useful? As opposed to the unbiased compound library screens that seem to be the gold standard. I may or may not be thinking on organizing a symposium to address this question.

Asking the void: are there good examples of compounds that targeted both host and pathogen enzymes (homologues) that were successfully modified/optimized to be selective for the pathogen’s enzyme?
