💡 Introducing TockyDevelopment: Our multidimensional toolkit encapsulates all methodologies and algorithms. It enhances understanding of T-cell development and broadens applications across various cell types, enabling experimental measurement of time-dependent trajectories in developing cells.

🐁 Case Study: Our toolkit excels in studying thymic T-cells from Nr4a3-Tocky mice. By analyzing Timer progression alongside multidimensional marker profiles, we decipher T-cell development like never before. By deleting Bim in Nr4a3-Tocky mice, we highlight time-dependent cell death.

🔄 Furthermore, we've developed the Dijkstra-Tocky pathfinding algorithm, a novel method using time-encoded data to trace probable paths of cell differentiation. By considering Timer progression and multidimensional marker profiles, we gain unprecedented insights into cell differentiation processes.

🛠️ To improve high-dimensional clustering, our novel PCA-based algorithm distinguishes genuine marker expressions from background noise, improving clustering accuracy and preserving developmental pathways crucial for detailed trajectory analysis.

📊 To solve the problem, Canonical Correspondence Analysis (CCA) has been adapted for high-dimensional flow cytometric Fluorescent Timer analysis! CCA now cross-analyzes Timer progression with multifaceted marker profiles, unveiling temporal trajectories in cellular development.

🧬 At the core is our Tocky technology, utilizing a Fluorescent Timer reporter gene to track time since a cell receives a developmental cue. By combining high-dimensional marker profiles with Timer fluorescence data, we decipher time-dependent changes during cell development.

Background: Capturing dynamic processes of cell development at the single-cell level in vivo is challenging due to the stochastic nature of signal transduction and lack of adequate methodologies. We've tackled these hurdles in both experimental and computational analysis.
