Quantum Computing Calculations of Protein–Ligand Binding Energies Using Decomposition Methods on Simulated and Real Quantum Hardware
We analyzed the performance of quantum algorithms for studying protein–ligand systems, using both simulator and real hardware. To achieve this, we selected thrombin and 5 ligands as a test case and employed a decomposition strategy using density matrix embedded theory, dividing the ligand systems into three fragments each. First, the energy of one of the fragments was calculated using the Variational Quantum Eigensolver (VQE) using a state vector simulator, while the energy of the remaining fragments was calculated using Couple Cluster Singles and Doubles (CCSD), with the protein treated as point charges located at the respective atomic sites. Through these noiseless simulations, we evaluated the results under ideal conditions using the state-of-the-art Unitary CCSD ansatz to validate the efficacy of the strategy in a controlled setting without quantum noise. Subsequently, we evaluated the impact of approximations in the ansatz, optimizer, and active space size, which are necessary to decrease the computational cost, in order to target real hardware during the current Noisy Intermediate-Scale Quantum era. Finally, we performed the VQE calculations using a superconducting quantum computer developed by the RIKEN RQC–Fujitsu Collaboration Center and also analyzed the noise effect through simulations. The results demonstrated that the use of quantum algorithms can enhance the binding energy correlation value, offering potential applications in the workflow of computer-aided drug design.

The spatial proteome of the Plasmodium falciparum schizont illuminates the composition and evolutionary trajectories of its organelles | Nature Communications
Malaria is caused by apicomplexan parasites of the genus Plasmodium, with all malaria symptoms and pathology caused by parasite stages that develop within, or transit between, host erythrocytes. The ability of Plasmodium cells to parasitise erythrocytes depends on distinctive intracellular compartments associated with invasion, as well as the development of unique cellular niches within the infected host cell. However, our understanding of the biology of the malaria parasite is limited by the fact that a large proportion of the parasite’s proteome has no known cellular location or function. To address this problem, we have generated comprehensive high-resolution maps of protein subcellular localisation for the invasive stage of the erythrocytic life cycle of Plasmodium falciparum, the major cause of malaria mortality. Systematic design of combination therapy by targeting master regulators of coexisting diffuse midline glioma cell states | Nature Genetics
Intratumor heterogeneity fundamentally challenges cancer treatment, as coexisting, molecularly distinct cell states with non-overlapping drug sensitivities can drive therapeutic resistance. We establish and validate a generalizable, network-based framework to systematically identify combination therapies targeting complementary tumor cell states. Applied to diffuse midline glioma (DMG)—a universally fatal pediatric malignancy—this approach identified master regulator protein dependencies in seven coexisting cell states, confirmed by pooled CRISPR–Cas9 assays. Perturbational transcriptional profiles for 372 clinically relevant drugs prioritized candidates predicted to invert state-specific master regulator activity. State-selective drug sensitivity was validated for eight out of nine (89%) drugs in vivo, including avapritinib, ruxolitinib and larotrectinib. ChatGPT Health performance in a structured test of triage recommendations | Nature Medicine
ChatGPT Health was launched in January 2026 as OpenAI’s consumer health tool and has reached millions of users. Here we conducted a structured stress test of triage recommendations using 60 clinician-authored vignettes across 21 clinical domains under 16 factorial conditions, yielding 960 total responses. Performance followed an inverted U-shaped pattern, with the most dangerous failures concentrated at clinical extremes—nonurgent presentations (35%) and emergency conditions (48%). Among gold-standard emergencies, the system undertriaged 52% of cases, directing patients with diabetic ketoacidosis or impending respiratory failure to 24–48 h evaluation rather than the emergency department, while correctly triaging classical emergencies such as stroke and anaphylaxis.
