More on the reservoir. The AGING cohort in Argentina (defined as 50 or older!!!) looked at total DNA and pro-inflammatory cytokine profiles longitudinally over 1 year. No changes but a short follow up time. I’d be interested in following this cohort over time w longer follow up analyses #IAS2025

This #IAS2025 poster looked at the #HIV reservoir in the Rakai cohort Uganda, measured w QVOA. As we know, QVOA < total HIV DNA. Interestingly QVOA doubled over 5 years for women in the cohort pre- and post-menopause. More evidence that female sex hormones impact reservoir. IPDA data coming

Joseph Eron - LEN plus 3bnc117 and 101074 (bought by Gilead and now “TAB and ZAB”). I think this was a TREATMENT study. 6 monthly dosing of LEN TAB and ZAB. 3 virological failures (but Joseph using the wording of viral rebound as if the participants underwent treatment interruption).

Alright that’s me done for #CROI2025 I am saturated with science. What a great meeting! At a time of disruption, despair and disinformation, it’s great to come together with colleagues and peers to progress HIV science, no matter what barriers we face. If off to the Castro for drink ✌️

Fascinating study where people with #HIV and cancer who required stem cell transplants received stem cells that had 3 HIV resistance genes added to them. These hung around for up to 2 yrs post transplant. Approx 30% mortality, one person elected to pause treatment and rebounded at week 7.

The TRAILBLAZER study randomised 30 people w #HIV to receive an infusion of CD4 cells edited to cut out CCR5 gene (thus protecting cells from being infected with CCR5 virus) or unedited cells. Didn’t work unfortunately. Only 2% of circulating cells edited and no reduction in reservoir. No ATI

Last few observations from #CROI2025 - there is so much science here & I’m overwhelmed at the quality & content of presentations #HIVcure from stem cell transplants was an insane breakthrough in the field, we have two cure POSTERS. The original Düsseldorf patient data was also a CROI2019 poster!

In the final talk, Lydie Trautmann discusses the importance of T cell responses in the brain and CNS and the role this may play in #HIVcure trials

Final symposium of #CROI. Prof Melissa Churchill from RMIT is talking about what we know about #HIVreservoirs in the brain and central nervous system. A lot of different groups have found evidence of intact and replication competent virus in brain tissue and various cell types.

Argh!!! #CROI2025 is practically over and the poster hall is being packed up! How? I feel like I’ve had a chance to look at hardly any! With 1200 posters at the conference it’s impossible to absorb it all, but there are a few that interested me. I’ll have to find them online later.

One observation from me during the hot new #HIV therapies session at #CROI2025. Others have been posting abt the data showing options for LA CAB/bNAb, ISL/DOR, and CAB/RPV in Africa but my hot take is: Look at the impressive % of women in these trials! 58% in CARES 21 & 40% in the ISL/DOR studies

News from #CROI2025: Two more people appear to be free of HIV after stem cell transplants for cancer treatment. If these men remain in remission, they will be the 8th & 9th known cases of a functional cure after the procedure. www.poz.com/article/two-...
